Marriage
She married George Ayliffe (later Sir George Ayliffe) of Brinkworth, Wiltshire.[1][2][3]
The Inquisition Post Mortem of her father in law John Ayliffe referred to an indenture of 02 Nov 1610 between John Ayliffe and his son George Ayliffe of the one part and John St John of Lydiard Tregoze and Anne St John his sister of the other part. John Ayliffe was to transfer 60 acres of meadow and 200 acres of pasture of the manor of Grittenham, Wiltshire, to his son George for his life, and after his decease to the use of Anne St John for her life, for her jointure.[4]

Death
Anne had died by May 1634 as her husband George Ayliffe married second Martha Blount on 20 May 1634.[11]
Anne was buried in the church of Lydiard Tregoze, Wiltshire. In the will of Sir George Ayliffe of Rabson, WIltshire, the will made on 22 May 1640 and proved on 30 Mar 1647[5] his will was "to be buried in the vault in Lydyard Church by the bodie of most deare wife there interred alredy which was one of her last requests to me." As his second wife was still living he was referring to his first wife, Anne.

"The triptych was erected, although not exactly in the form it takes today, in 1615 by Sir John to commemorate his parents. The two central doors open to reveal the life-size painted figures of Sir John St. John (d. 1594) and his wife Lucy Hungerford (d. 1598) kneeling upon a tomb. To the left stands Sir John, the younger (d. 1648), the erecter of the monument, and his first wife Anne Leighton; to the right are his six sisters with shields of arms at their feet. When the central leaves are closed, their outer surfaces and the panels and leaves flanking them bear an elaborate display of painted heraldic genealogical tables of the St. John family. These tables were originally based upon the genealogical work of Sir Richard St. George, Clarenceux King of Arms, and Sir John's uncle by marriage, but many additions were made subsequently".[12]
